What lengths are available for Multiplexed Gene Fragments (MGFs)?


MGFs range from 301–500 nucleotides, including flanking sequences used for PCR amplification. Flanks must be 20–25 bp, allowing up to 460 bp of variable coding region. Fragment length variation within a pool should not exceed 80 bp.

What are Twist’s Multiplexed Gene Fragments?


Twist MGFs are pooled double-stranded DNA fragments (301–500 bp) designed for high-throughput applications like prime editing, CRISPR functional screens, peptide/protein engineering, antibody discovery, and MPRA. This pooled format reduces cost per variant while enabling large-scale experiments.

How are Multiplexed Gene Fragments produced?


Twist uses phosphoramidite chemistry on a miniaturized, high-throughput platform. This reduces reaction volumes and increases throughput, enabling pools up to 500 nt. Fragments are amplified into dsDNA via an optimized workflow for downstream applications.

Why does Twist amplify the pools?


Amplification ensures reliable dsDNA output and mitigates challenges associated with PCR amplification of synthesized material, improving consistency across experiments.

How should I upload my antibody sequences?
  • Remove signal leader peptides if using pTwist vectors with Twist-recommended signal leader.

  • Variable regions should not end with CH1 sequences.

How long does custom vector onboarding take?


Onboarding takes 10–15 business days from the assigned start date (once vector aliquot and completed form are received). Any unexpected issues are communicated promptly with a revised completion date.

What is the shelf life of Gene Fragments and Clonal Genes?
  • Dried DNA: RT 3 months, 4°C 12 months, -20°C 24 months, -80°C long-term.

  • Resuspended DNA: Store in TE or elution buffer, not water. Use aliquots to minimize freeze-thaw cycles.

How much DNA is delivered?
  • Gene Fragments: 100–1,000 ng per fragment.

  • Clonal Genes: Microprep 50 ng–2 µg, Miniprep 2–10 µg, Midiprep 10–100 µg, Maxiprep 100 µg–1 mg. Yield may vary based on sequence or vector; low-yield fragments still ship at no charge.

How is synthesis quality tested?
  • Clonal Genes: NGS-verified.

  • Gene Fragments: Not NGS-verified; sequencing 3–6 colonies (depending on fragment size) achieves 99% confidence in a perfect clone.

What concentration is delivered per oligo in a pool?


Each oligo is delivered at >0.2 fmol per oligo.
